    Abstract: CDKL5缺失疾患由CDKL5基因突變引起。患者多在出生後六週內即頻繁發生癲癇。我們先前發現CDKL5缺失小鼠於出生後12天出現癲癇的徵狀,但隨即恢復正常。透過全基因組RNA序列分析,我們發現CDKL5缺失導致新生小鼠海馬迴中抑制性神經傳導相關基因的表現量下調,而離乳前小鼠皮質中GABA傳導相關基因的表現量卻上調。本計畫將首先確認CDKL5缺失是否透過海馬迴中抑制性神經傳導的減少而增加了出生後早期的神經興奮性(Aim 1)。然後探討CDKL5缺失小鼠在離乳前神經興奮性回復正常是否肇因於皮質中GABA傳導的提升(Aim2)。本計畫之研究成果將有助於發展不同年齡適用的療法來改善CDD的早發性癲癇。
