    Abstract: CDKL5缺失疾患由CDKL5基因突變引起,患者多在出生後六週內即頻繁發生癲癇,影響神經發育。本計劃之先導實驗發現CDKL5缺失小鼠於出生後12天出現癲癇的徵狀,但隨即恢復正常。而P7幼鼠之CDKL5缺失增加神經元興奮頻率且下調與切線遷移相關的基因轉錄。我們因此假設:CDKL5會隨年紀調節基因表現,以維持正常的切線遷移和神經興奮性。本計畫將首先確認在不同年紀下CDKL5缺失對神經興奮性及基因表現的影響 (Aim1),然後將探討CDKL5缺失是否影響發育中不同時期中間神經元的切線遷移 (Aim2)。本研究之結果將有助於評估未來是否以中間神經元切線遷移的調控因子為標的,發展早發性癲癇的治療。
